Semax is a synthetic heptapeptide (Met-Glu-His-Phe-Pro-Gly-Pro) consisting of the ACTH(4-7) fragment extended with the C-terminal tripeptide Pro-Gly-Pro, which confers metabolic stability. It has no hormonal (corticotropic) activity. Developed at the Institute of Molecular Genetics in Moscow, it is used clinically in Russia in ischaemic stroke.
Genome-wide transcriptional studies in rat focal cerebral ischaemia show Semax markedly alters expression of immune- and vascular-system genes, with over half of the affected genes relating to immune response — proposed as the basis of its neuroprotective action 12.
