PT-141 (bremelanotide) is the deamidated metabolite of Melanotan II, a cyclic heptapeptide melanocortin agonist with activity at the MC4 receptor, which governs central sexual-arousal pathways.
In the two phase 3 RECONNECT trials in premenopausal women with hypoactive sexual desire disorder (n = 1,267), bremelanotide significantly improved sexual desire and reduced associated distress versus placebo 1; a 52-week open-label extension supported long-term safety 2. It was approved by the FDA in 2019.
