Melanotan II is a cyclic lactam-bridged heptapeptide analogue of α-MSH(4-10) with superpotent melanotropic activity across melanocortin receptors (MC1, MC3, MC4, MC5).
In a pilot phase 1 study, five low subcutaneous doses produced measurable skin darkening in healthy volunteers 1; a subsequent placebo-controlled study observed erectile responses in men with psychogenic erectile dysfunction, leading to the development of the MC4R-focused metabolite bremelanotide 2.
