PNC-27 is a 32-residue peptide combining the p53 HDM-2-binding domain (residues 12–26) with a membrane-residency (penetratin) sequence. Cancer cells express HDM-2 in their plasma membrane; PNC-27 binds it and forms transmembrane pores, causing rapid necrosis of the cancer cell while sparing normal cells lacking membrane HDM-2.

This mechanism — termed 'poptosis' — has been characterised across multiple cancer-cell lines 12; recent work shows PNC-27 kills cervical cancer cells but not normal cervical cells 3.
