FOXO4-DRI is a D-retro-inverso peptide that competitively interferes with the interaction between FOXO4 and p53 in senescent cells, releasing p53 to the cytoplasm and inducing selective apoptosis of those cells.
In the defining Cell paper, FOXO4-DRI neutralised senescent cells in fast-aging XpdTTD/TTD mice and naturally aged mice, restoring fitness, fur density and renal function 1. It is one of the foundational compounds of senolytic research.
