PE-22-28 is a seven-residue analogue of spadin, a natural peptide derived from sortilin propeptide that blocks the TREK-1 background potassium channel. TREK-1 inhibition is a validated antidepressant mechanism in rodent models.

Shortened spadin analogues including PE-22-28 displayed stronger TREK-1 inhibition, greater in-vivo stability and faster antidepressant-like activity than the parent peptide, with effects evident after four days of treatment 1.
