MOTS-c is a 16-amino-acid peptide encoded within the mitochondrial 12S rRNA gene — one of the 'mitochondrial-derived peptides' that act as retrograde signals from mitochondria to the nucleus. It targets the folate–methionine cycle, raising AICAR and activating AMPK in skeletal muscle.
In the defining study, MOTS-c administration in mice prevented diet-induced obesity and age-dependent insulin resistance, enhancing glucose utilisation and metabolic flexibility 1.
