Humanin is a 24-amino-acid peptide encoded in the mitochondrial 16S rRNA gene. It was discovered as a rescue factor in a screen of surviving neurons from an Alzheimer's-disease brain.

In the discovery study, humanin abolished neuronal cell death induced by a wide spectrum of familial Alzheimer's-disease genes and amyloid-β, establishing it as the first mitochondrial-derived cytoprotective peptide 1.
