Melanotan I is [Nle4, D-Phe7]-α-MSH, a linear 13-residue analogue of α-melanocyte-stimulating hormone that is far more potent and stable than the native hormone. It activates the melanocortin-1 receptor on melanocytes, stimulating eumelanin synthesis.

As afamelanotide, it is approved for erythropoietic protoporphyria: in two phase 3 trials it increased pain-free sun exposure and improved quality of life 1.
