IGF-1 LR3 is a recombinant analogue of insulin-like growth factor 1 with an Arg substitution at position 3 and a 13-residue N-terminal extension. These changes dramatically reduce affinity for IGF-binding proteins, increasing free, biologically active concentration 1.

In rat studies, IGF-1 variants with reduced binding-protein affinity were more anabolic than native IGF-1 — increasing body weight and nitrogen retention at lower doses 2. Structural characterisation by NMR confirmed the analogue retains the native IGF-1 fold 3.
