CJC-1295 with DAC adds a maleimidoproprionic-acid linker (the Drug Affinity Complex) to the tetra-substituted GRF(1-29) backbone. The linker covalently binds circulating albumin, extending the half-life from minutes to roughly six to eight days.
In healthy adults, single CJC-1295 doses increased plasma GH 2- to 10-fold for six days or more and IGF-1 1.5- to 3-fold for 9 to 11 days; repeated weekly or bi-weekly dosing maintained the elevation with an estimated half-life of 5.8–8.1 days 1.
