GHRP-6 (His-D-Trp-Ala-Trp-D-Phe-Lys-NH2) was the first synthetic peptide shown to release GH through a receptor distinct from the GHRH receptor 12. The search for its endogenous ligand ultimately led to the discovery of ghrelin and the GHS-R1a receptor 3.

It stimulates GH release in vitro and in vivo and is also associated with increased appetite via ghrelin-receptor signalling.
